I was listening to a reference song for a customer and I noticed this overtone at the end of the song to go along with the Power Chord. I thought it might be an organ, or some other instrument, but can't hear an organ-or any other instrument with this sound- during the track at all. So, I'm wondering if it's a room overtone, or if the overtone was added through some other way.
Any thoughts would be appreciated.

You're hearing the high 3rd over the chord?
There's a powerchord and a barre chord there seperate, to my ears, and the tone you're hearing is just an overtone of that barrechord.

I could almost swear I heard some amplifier distortion elevating the amplitude of 1st and 2nd order harmonics.
